Collagen for pharma and drug delivery

A delivery system succeeds or fails on release. Too fast and the dose is wasted before it does anything. Too slow and the therapy never reaches concentration. Collagen has been used as a carrier for decades because it degrades enzymatically at a rate the body sets, holds high payloads of water soluble and poorly soluble molecules, and leaves behind peptides the tissue already knows how to clear.

What we supply

The reason more programmes do not use it is supply. A carrier material is on the critical path of a regulatory submission, and the questions asked of it are unforgiving. What is the molecular weight distribution, what is the endotoxin level, where did the animal tissue come from, what happens to all of it batch to batch across three years of clinical work. A supplier who cannot answer in writing becomes a risk the formulation team carries alone.

Alpstein supplies bovine type I collagen as a pharmaceutical carrier in the formats formulation work needs. Soluble atelocollagen solution for injectable gels, in situ forming depots and coating suspensions. Soluble atelocollagen powder for reconstitution and for lyophilised presentations. Fibrous atelocollagen paste and powder for implantable depots, sponges loaded with an active, and sustained release matrices. Sheet for local delivery at a surgical site. Sterile grades throughout where the route demands it.

The material ends up in injectable depots for local and sustained release, implantable sponges carrying an antibiotic or a growth factor, microsphere carriers that hold active ingredients stable at room temperature after ultra low temperature vacuum drying, ocular inserts and shields, periodontal and intra articular delivery, aesthetic injectables combining collagen with hyaluronic acid, vitamin C, PRP or botulinum toxin, and coated devices where the drug sits in a collagen layer on a metal or polymer surface.

What Alpstein brings to the formulation

01

Release rate follows fragment length and crosslink density. Alpstein defines fragment length inside a closed process across multiple centrifuge stations, so the release profile you characterise in early formulation is the one you see at clinical scale.

02

The triple helix arrives intact at approximately 300 kDa, which keeps enzymatic degradation predictable and keeps the carrier from collapsing into a burst release you did not design.

03

Purity is specified where regulators look. Total protein above 98 percent, non-collagen protein below 1 percent, endotoxin below 0.5 EU per mL, pH 6.5 plus or minus 0.5. For a parenteral route, every one of those lines belongs in your specification and each one is already there.

04

Animal sourcing is restricted to WOAH negligible risk countries, with ISO 13485:2016 and ISO 22442 parts 1, 2 and 3 applied through manufacture, unbroken cold chain in transit and traceability to the herd. The viral and TSE safety section of a submission is the section that stalls programmes, so the documentation is prepared for that conversation rather than assembled when it arrives.

05

Independent purity and identity data comes from Intertek. Mechanical and structural characterisation comes from Cambridge and Oxford. Third party analysis carries further in a regulatory review than a supplier’s own certificate.

06

Development support is available where a formulation needs the carrier changed rather than chosen. Loading method, porosity, degradation window, and compatibility with the active.

Why Alpstein

Release tracks tissue activity rather than a fixed dissolution curve.

Enzymatic degradation the body regulates.

The release profile characterised in development survives the move to scale.

Defined fragment length.

Soluble and poorly soluble actives both load into the matrix.

High payload capacity.

Microsphere collagen carriers hold activity without cold storage at the point of use.

Room temperature stability through lyophilisation.

Protein content, non-collagen protein, endotoxin and pH all sit in the incoming specification.

Parenteral grade purity specified.

Injectable and implantable routes served without an added processing step.

Sterile supply.

Works alongside hyaluronic acid, vitamins, PRP, antibiotics and growth factors.

Compatible with combination products.

Prepared for the section of a submission that stalls programmes.

Full viral and TSE safety documentation.

Intertek for purity, Cambridge and Oxford for structure and mechanics.

Independent characterisation.

A carrier on a ten year product life needs a supplier with the same horizon.

Long term supply from a Swiss company.

Loading, porosity and degradation window tuned to the active.

Formulation development support.

The release profile you characterise is the release profile you keep.
